With how low hanging fruit that joke is and how much crossover I assume exists between Star Trek fans and Renaissance Fair attendees, that probably happened the first Renaissance Fair after Star Trek got popular.
Probably even happened at fairs during the actual Renaissance, centuries before Star Trek.
I don't think Star Trek has really taken off yet back then.

Check with the rule for the fair. Some fairs do not want people who arent staff dressing up in costumes because it can lead to people impersonating staff.
If they have no such rule do what you want and have fun.

If you're going as patron, you can wear whatever you want. I've been going to the local Ren Fest for almost 20 years and I've seen all kinds of crazy outfits-- Star Trek characters, Bill and Ted, LotR, Marvel characters, etc. Only the people that actually work there have to adhere to a dress code.
I would imagine that it will depend on how .. into it the fair is. The one that hits my area, we civilians aren't dressing up at all. It also seems like people really stretch the meaning of "Renaissance" as people are in medieval garb, which isn't Renaissance, and you've got people dressed as Jack Sparrow-esque pirates. YEah Roman's would be way out of Renaissance times, but so are half of the other costumes. so why not.
I gotta say I really like the gag of someone dressing in a starfleet uniform having a tricorder and walking around a renaissance fair.
I know nothing about this scene, but the one time I went to one of these events, I, too, noticed that “people really stretch the meaning of‘Renaissance’.”
I do also think they had a separate steampunk day, though.
