Yeah - I was doing some additional digging and I’m pretty sure it’s cucumber mosaic virus.

The biggest two changes lately were (A) a ton of rain over last few weeks, maybe 6inches+. So could be nutrient deficiency? And (B) noticed a bunch of insects on the bottoms of the leaves and on fruit that arrived in last few days. I thought they were mostly potato aphids but definitely could have been a mix. Image attached of some of the bugs. Definitely more on new leaves (and a couple of nodes down) than old leaves. Little suckers on previously trimmed areas don’t seem effected and have normal leaves.

UNC Pediatrics at Southpoint had them as of a few months ago.

I really enjoyed the montage where they fast forward and summarize how quickly they discovered new things about the language and made new tools. That’s is the excitement of new science!!

Not completely true - see the Gizmodo link below. The first link for OP goes through the saga. Eufys practices show that you cannot trust them as a company even when you’re local only. I think the only way it might be okay to setup is through HomeKit to avoid any account with Eufy … but I still do not trust them and have stopped using all my Eufy cameras.

Any tips for bossing with bone spear?[Question] Builds | Skills | Items

I’m running the wowhead bone spear build/paragon and despite folks claiming BS is good for bossing, I always felt that it shines way more in NMD clearing as compared to the bossing. Right now I can usually casually clear through a NM95-100 until the boss but if I don’t have at least two lives left for boss chances are I’m not completing the dungeon. Any tips or optimization you all have here?

Definitely still a good number of research roles here too (especially in gene therapy and editing, viral delivery, muscle bio, and antivirals) but less that I see fall into the open market. What type of area are you looking for roles in?

Surprised to not see stolen vigor here - any thoughts on that front?

Middle of the well is fine as long as your add it drop by drop slowly. Definitely a density issue - usually use about 4-4.5M cells per 10cm dish so feel free to use that as a scaling ratio for whatever format you use. Higher density will also help keep the cells happier and stuck on the plate after transfection (with more cell-cell contacts to stabilize).

-What was cell confluence like before transfection? 293s should be pretty confluent before transfection (90% ish) - way more than even the second image. If under confluent you might have nuked them. I would say even the second image doesn’t look like a good set of cells

-They also are sometimes a pain to keep evenly distributed in the wells and if you see more cells come off in the center it’s usually a sign the cells settled a bit too much in the center (try a figure 8 with the plate).

-did you rocket the transfection reagent into the wells or gently add as others have mentioned

-how much DNA did you add? 293Ts are super hardy to dna amount compared to other cels but you might have added too much DNA or added really “dirty” dna preps to the cells that killed them.

Automations not working consistently?

Hi all - starting to go a bit crazy trying to figure this out but wanted to see if anyone here could point me in the right direction. I’ve gotten a bunch of simple automatons up and running (eg. Turn on exterior zwave plug when it’s 4:45, turn on Lutron lights when contact sensor on door opens) but seemingly randomly there are days or triggers where the automations run but the associated actions never happen. Come back a trigger or two later and it works fine randomly.

Right now my setup is: -Zooz 700 plus s2 zwave USB (in cabinet - not sure if that’s killing the zwave signal?) -Zooz 700 zwave plus outlet switch -Yale assure zwave locks -Lutron caseta lights/fans driven by a Lutron Pro hub -Ring door sensors -a few Kasa bulbs -One set of Phillips hue bulbs and motion sensors

At first I thought it could be on the Lutron end since most automations are using light to trigger - sometimes the various switches in the dashboard don’t update properly with the current light status or aren’t responsive to manual commands. I also had seen that some folks using the Lutron Pro hub had some issues with connectivity with the standard Lutron integration when a fan controller was connected and had written some code to make that cleaner. However, even the automations that rely on the Zooz Zwave outlet won’t trigger randomly and the outlet doesn’t seem to be available to HA correctly at all times. Could this just be a zwave network issue combined with the Lutron issue? However, never had issues that I could tell with the Ring door sensor triggering and the Phillips hue bulbs have never really had any issues with the motion trigger automation I setup. Any thoughts about how to start diagnosing these types of issues if it is a zwave mesh issue?
