Obligatory not peter. It's a stupid throw-away joke. But there's a lot than can be read into it.
First, he's a Guardsman turned cow, a Cow Guard, or if we're going to call them Wards instead of Guards... He's a Cow Ward...
Next, he's a cow wanting to go home. "Until the Cows go home" is a saying. Nothing really related to this, but it explains why the cow wants to go home.
Finally, the point of the joke isn't either of the above. It's to point out the incongruity of the villainess treating this as an acceptable demand, and allowing him to leave, in the midst of her attempt to kill the protagonist. Incongruity is humorous, that's all.
Lastly of all, every other guard is totally fine to keep working
I feel like like noting the source material is a movie that can be summed up as "buddy comedy where guy turns into a llama, because Disney plots weren't cowards in TYOOL 2000", like that might be the final disconnect, because when you put it that way it sounds almost.......silly
But that would mean someone hasn't seen Emperor's New Groove and that's so sad

They're not female. There's no utters. There are plenty of instances of bulls without horns sharing the same coat as their steer counterparts. Considering the entire movie is without a script, it's most likely just a joke amongst the cast and animators. The entire movie is a Disney shitpost where whatever someone wanted in got in.

While that was just said, I’d like to mention that horns aren’t a male only thing in cows and bulls. It’s not like deer with antlers, where only the males have them. Similarly to goats, both male and female cattle have horns.

Yeah it’s a pretty crazy story behind this movie, I can’t remember the name but there is a documentary about its creation. Basically the artist Sting was brought in early to write music for it then Disney destroyed the budget because of how bad their last musical epics did. Brought in a new director and scraped the script so they just kinda wrote it on the fly lol
That’s why the music in the film is so epic feeling for a lighthearted comedy as the film is but also makes it feel so much better.

The context of this scene is that Kuzco, the Llama, is an Incan emperor who was betrayed by his former assistant, Yzma, the purple woman. Her plan was to poison him, but in a mishap involving her various potions, she just turns him into a llama. By this point, Kuzco has made it back to his palace but he needs to find the potion that will turn him human, that of which is mixed up with all sorts of other transforming potions. Since these guards are sent after him, he uses the potions against them and they're all turned into different animals as Kuzco escapes. Yzma sends them after him again, but the cow objects noting that he is a cow and now wants to go home and is properly excused before every other guard proudly charges forth after bring asked if anyone else wishes to leave. The joke is the subversion of expectations, with just one of the guards deciding to go home despite everyone else being an animal. This subversion is doubled upon when they have the cruel, normally unforgiving villain excuses him without issue before asking anyone else if they wish to leave. The joke is the subversion that she's a genuinely good boss compared to Cuzco, when asked if anyone else wants to leave, they said no and carried on with their jobs which is also humorous.

This is originally from a scene from the Disney movie Emperor's New Groove. The Guards get turned into animals because magical potions were thrown at them. One of them asks if they can go home. This is funny cause there's a bit of surrealism to it. This isn't in the meme but the other guards are fine with being animals and still do their job after this.

Nah, cows are gender-loaded so it's a fair guess, and they said they weren't sure. It's important to go "is this transphobia" even if the answer is "no", too, because then we understand it better.
